The curtain opens on a nostalgic flashback to when young Branch (Alan Kim) was part of the boy band BroZone, alongside his brothers John Dory (Eric Andre), Floyd (Troye Sivan), Spruce (Daveed Diggs), and Clay (Kid Cudi). Under the moniker Bitty B, they were poised to take the stage by storm, but Branch's nerves got the better of him. His trusted brother and confidant, Floyd, reassured him that everything would be okay, and with a collective blast of harmonious energy, BroZone was ready to shatter diamonds with their rendition of "Perfect Harmony". However, John Dory's showboating antics sent the performance spiraling out of control, resulting in a tangled mess of brothers as they tumbled off the stage. The ensuing fallout saw the siblings part ways, with Floyd leaving Branch in the care of their grandmother, promising to return for him.
In the present, an adult Branch (Justin Timberlake) harbors deep-seated resentment towards his brothers for abandoning him. Meanwhile, Poppy (Anna Kendrick) stumbles upon Branch's nostalgic reverie, oblivious to his former life as a member of BroZone. The scene shifts as King Gristle (Christopher Mintz-Plasse) and Bridget (Zooey Deschanel) prepare to tie the knot in a lavish ceremony ("Let's Get Married").
Just as the wedding festivities are about to get underway, John Dory makes a surprise appearance, bringing with him a crucial message from his brother Floyd. Trapped on Mount Rageous by the insufferable pop duo Velvet and Veneer (Amy Schumer and Andrew Rannells), Floyd is being held captive in a diamond perfume bottle that extracts his Troll essence to amplify their lackluster vocals. John implores Branch to rally the rest of his brothers to stage a rescue mission, but Branch is hesitant about reuniting with his former bandmates. Poppy's unwavering enthusiasm and Branch's sense of duty eventually win out, and they embark on an adventure to save Floyd.
As Gristle and Bridget exchange their vows in the presence of Miss Maxine (RuPaul), the stage is set for a thrilling quest that will require Branch and his newfound partner-in-crime, Poppy, to put aside their differences and join forces with the estranged members of BroZone.
As Velvet and Veneer ( ) wrap up their music video shoot for "Watch Me Work" and a candid interview, they depart from their beleaguered assistant Crimp (Zosia Mamet) to pilfer more of Floyd's essence. However, with each successive extraction, the once-lively Floyd succumbs further to the ravages of exploitation. Veneer is torn between abandoning his newfound fame and wealth, and the moral imperative to free Floyd from his prison. The dichotomy weighs heavily on her conscience as they prepare to depart.
Meanwhile, Poppy and Branch embark on a fantastical journey aboard Rhonda, a gigantic insect masquerading as a bus, accompanied by John Dory and an uninvited Tiny Diamond (Kenan Thompson). Their destination is Vacay Island, where Muppet-esque creatures dwell in harmony. The trio's quest is to locate Spruce, now rebranded as Bruce, following his marriage to Brandi (Patti Harrison) and the birth of their brood of over a dozen children. As they reunite with Spruce, the tension between him and John Dory, perceived as the architect of the band's demise, is palpable.
Upon learning about Spruce's new life, Poppy and Branch are treated to an impromptu performance by Spruce and his brothers in their boy band heyday. However, the real mission begins when they receive intel that Floyd is in dire need of rescue. Brandi offers her support, but with a caveat: Bruce must complete the task on his own for a month upon their return.
Crimp's latest invention, a device capable of crafting smoothies and essence sprays from fruits, takes an ominous turn when Velvet employs it to absorb Floyd's essence using the diamond prison, leaving Crimp horrified by her actions.
As Tiny Diamond presses the "hustle" button on Rhonda, the Trolls find themselves transported through a psychedelic portal to a secret golf course, where they encounter Clay. Their meeting with Viva (Camila Cabello), an über-enthusiastic pink Troll, reveals that she is Poppy's long-lost sister, sharing King Peppy (Walt Dohrn) as their father. The revelation sparks excitement in Poppy ("It Takes Two").
However, as the brothers share their mission with Clay, Poppy and Branch separately discover that Viva was separated from her family during the Trolls' and Bergens' tumultuous past. When they attempt to depart, Viva's determination to protect her land and its inhabitants from what she perceives as evil Bergens becomes apparent. Despite Poppy's attempts to reason with her sister, Viva remains resolute in her decision not to join their quest.
As a desperate ploy, Floyd feigns his own demise in a bid to gain his freedom from Velvet's clutches. However, Veneer's watchful eye catches him in the act, and despite Floyd's attempts to appeal to his better nature, he ultimately proves no match for Velvet's ruthless determination. Forced to retreat, Floyd finds himself captive once more.
Meanwhile, Clay joins forces with Poppy and her companions as they converge on Mount Rageous, their journey marked by a series of explosive confrontations sparked by John Dory's provocative antics. As the group begins to fracture along its seams, Branch's pent-up frustration boils over, prompting him to storm off in a fit of pique. However, it is Poppy who ultimately prevails upon her friend, coaxing him into forgiveness and reconciliation.
As Gristle and Bridget embark on their honeymoon, their motorcycle escapades take them to the unlikeliest of destinations: a golf course. Here, they fall prey to Viva's Troll tribe, who capture the star-crossed lovers and prepare to dispatch them to a gruesome fate in a meat grinder. Undeterred, Gristle and Bridget opt for a romantic swan song, their ardent displays of affection momentarily captivating their captors. It is only when Viva recognizes Bridget from Poppy's scrapbook that she begins to question her own motivations, ultimately deciding to join forces with the star-crossed pair.
As the Trolls attempt to infiltrate Velvet and Veneer's lair, they find themselves foiled by the villains, who capture John Dory, Clay, and Spruce, subjecting them to their essence-stealing devices. As the dust settles, Poppy, Branch, and Tiny set out to rescue Crimp, only to discover her imprisoned in a closet, awaiting her fate.
Rhonda's timely intervention allows Poppy, Branch, and Viva to confront Velvet and Veneer before a crowd of expectant onlookers at the Rage Dome. With a deft flourish, they expose the duo's true nature as charlatans, sending shockwaves through the gathering. As the chase is on across Mount Rageous, it is Viva, Gristle, and Bridget who arrive just in time to lend their support.
The brothers' combined efforts ultimately yield the perfect harmony, shattering the diamond prisons that had held John, Clay, and Spruce captive. As the Trolls return to their idyllic kingdom, Viva is overjoyed to be reunited with Peppy after an extended separation. The reunion is short-lived, however, as Branch takes center stage to reveal a stunning secret: he was part of not one, but multiple boy bands prior to BroZone. With a mischievous glint in his eye, he summons four fellow Trolls – Lance Bass, JC Chasez, Joey Fatone, and Chris Kirkpatrick – for an unforgettable musical spectacle that pays homage to the iconic *NSync.
