As Megamind (Megamind) donned the mantle of Metro City's protector, he found himself shouldering a new responsibility alongside his trusted ally Ol' Chum (Ol' Chum), formerly known as Minion. Together, they thwarted the nefarious plans of the Go-Fish gang, foiling their attempt to pilfer a priceless aquatic treasure. The citizens of Metro City rewarded Megamind's bravery by bestowing upon him the key to the city, solidifying his status as a champion. However, Chum began to feel overlooked and undervalued in their partnership, prompting him to seek out new opportunities. Megamind, sensing his friend's disillusionment, permitted Chum to explore other options.

Meanwhile, the Doom Syndicate, a notorious coterie of supervillains that Megamind had once been a part of, emerged from their prison cells with the intention of visiting their former associate, believing him to be still consumed by malevolent intentions. They embarked on a crime spree, challenging Megamind to prove his villainous nature by orchestrating a daring bank heist. However, Megamind's struggles were alleviated by Keiko Morita (Keiko Morita), his self-proclaimed biggest fan, who aided him in staging a convincing fake robbery.

As Chum found himself working at a dilapidated diner, Megamind continued to deceive his old cohorts into believing he was still harboring evil designs. He revealed an abandoned plan to launch Metro City to the Moon using rockets secretly planted beneath the city's surface. The Doom Syndicate prepared to put their scheme into action, but Megamind cleverly stalled them, aware that they would either launch the city or destroy it. Recognizing he couldn't vanquish the Syndicate alone, Megamind tracked down Chum at his remodeled diner and attempted to recruit him for his cause. However, upon realizing Chum's newfound sense of purpose, Megamind regretted letting his former sidekick slip away.

As the Doom Syndicate closed in on Megamind, they discovered his facade and overwhelmed him with their superior numbers. Feeling defeated and disheartened, Megamind retreated to an alleyway, only to be confronted by Roxanne (Roxanne) and Keiko, who reminded him that it's acceptable to seek help from others when needed. Chum soon reappeared at Megamind's side, as he acknowledged his former partner's true value as his trusted sidekick.

Together, the quartet of heroes – Megamind, Chum, Roxanne, and Keiko – joined forces to take on the Doom Syndicate, but ultimately failed to prevent the launch. However, they managed to redirect Metro City safely back to Earth. With the city's fate secured, Roxanne was elected as its new Mayor, while Keiko activated a newly designed hero signal, symbolizing Megamind's growth and acceptance of his role as part of a team.

In a mid-credits scene, the Doom Syndicate found themselves once again imprisoned, visited by none other than Megamind's former mentor, Machiavillain.
