Five years have elapsed since the events of the preceding film, and Greg Focker (played by) has been busy preparing for a milestone birthday celebration with his wife Pam - the twins Samantha and Henry are turning five. However, their joyous occasion is short-lived, as Greg's in-laws, Jack and Dina Byrnes, arrive unannounced, bearing news that sends shockwaves through the family.

It transpires that Jack has been diagnosed with a heart condition, which has left him feeling bitter about his daughter Debbie's divorce from her husband Bob. This development has led Jack to reevaluate his plans for the future of the Byrnes family, and he sees Greg as the perfect candidate to take over as head of the household. Despite Greg's reluctance, Jack bestows upon him the esteemed title of "The Godfocker", a moniker that seems more like a joke than a genuine honor.

As Greg struggles to come to terms with his new role, Jack's doubts and suspicions begin to simmer just below the surface. He becomes increasingly paranoid when he catches Greg consorting with Andi Garcia, a charming drug representative who doesn't seem to be afraid of flaunting her charms. The presence of Sustengo, an erectile dysfunction medication that Greg promotes as a side hustle, only serves to fuel Jack's concerns about his own marriage and virility.

Meanwhile, Bob, the ex-husband of Debbie Byrnes, shows up at a medical conference where Greg is promoting Sustengo. Bob shares with Greg the original plan to name him "The Bobfather", which leaves Greg feeling both relieved and uneasy. As tensions escalate between Greg and Jack, the latter becomes increasingly convinced that his wife Pam would be better off with her ex-fiancé Kevin Rawley, who he perceives as more financially stable and personally compatible.

In a desperate attempt to escape the chaos, Greg retreats to his and Pam's unfinished new home, only to find Andi waiting for him. She tries to lift his spirits with Chinese food and wine, but ultimately becomes too inebriated to distinguish reality from fantasy. Her aggressive advances on Greg are met with resistance, but not before Jack arrives at the scene, witnessing what he believes to be a compromising situation.

Disgusted and distraught, Jack departs the house, leaving Pam and Dina none the wiser about his discovery. The stage is set for a series of misunderstandings, misadventures, and comedic mishaps that will test the mettle of Greg's marriage and his ability to navigate the treacherous waters of family dynamics.

The tension at the twins' birthday party reaches a boiling point as Bernie (Greg's father) and Roz (Greg's mother) rejoin the family gathering the day after. However, the festivities are soon overshadowed by a surprise announcement from Bernie: he and Roz have decided to relocate to Chicago, with plans to settle just two houses away from Greg and Pam's new home. Jack and Dina (Greg's wife) are equally enthused about the prospect of being close to their grandchildren, sparking a debate between the couples.

As the credits roll, a glimpse into Jack's post-holiday routine reveals him browsing YouTube videos featuring Greg's public mockery during a Sustengo promotional speech – a lighthearted moment that belies the deeper connections forged among the family members throughout the year.
