As the milestone wedding date looms closer, Greg Focker (no actor name provided) and his betrothed Pam Byrnes decide it's time to bring their parents together under one roof. The unsuspecting families converge in Oyster Bay, New York, where they pick up Pam's father, retired CIA operative Jack Byrnes (played by Robert De Niro), her mother Dina, and her precocious one-year-old nephew, Little Jack Banks - the offspring of Bob and Debbie Banks. The patriarch of the Byrnes family, Jack, pilots his newly acquired RV to Miami, Florida, where they're set to meet Greg's eccentric parents, Bernie Focker (played by Dustin Hoffman), a former lawyer turned stay-at-home dad, and Roz, a sex therapist specializing in geriatric couples.

As the two families mingle, cracks begin to form between Jack and the Fockers due to their vastly different personalities and backgrounds. While Dina effortlessly connects with the free-spirited Fockers, Jack finds himself increasingly at odds with them. The tension is palpable as the Byrnes' cat, Jinx, and the Fockers' dog, Moses, engage in a high-stakes game of feline-canine tag that culminates with Jinx flushing Moses down the RV's toilet. Bernie Focker springs into action to rescue his beloved pet, destroying the toilet in the process.

Tensions continue to simmer as the two families clash over their vastly different approaches to life. Jack's back takes a turn for the worse during a game of touch football after Bernie Focker accidentally injures him with an errant tackle.

Pam confides in Greg that she's expecting, and they decide to keep the news under wraps until they're officially married. Meanwhile, Bernie Focker inadvertently reveals to their guests that Greg lost his virginity to their former housekeeper, Isabel Villalobos, 15 years prior. This bombshell sparks a chain reaction as Isabel's 15-year-old son, Jorge - who bears an uncanny resemblance to Greg - catches Jack's attention after he repairs the toilet in the RV.

As Roz, Bernie, and Dina vow to keep Pam's pregnancy under wraps from Jack, they're left to navigate the treacherous landscape of their own relationships. Greg is left to fend for himself as the lone babysitter of Little Jack, who's been raised via the strict Ferber method by his grandfather. Despite Jack's instructions to leave Little Jack to self-soothe, Greg finds it impossible to resist the toddler's cries and attempts to comfort him with humor and affection - inadvertently teaching him a rather... colorful vocabulary.

The situation reaches a boiling point when Jorge accidentally lets Little Jack out of his playpen. The mischievous tot turns on the TV, becomes fixated on Scarface, and proceeds to glue his hands to a bottle of rum while shouting "asshole" repeatedly. This chaos sparks an argument between Jack and the Fockers over their differing parenting styles, bringing the entire family's dynamics to a head.

As Jack's surveillance intensifies, he dispatches DNA tests to Greg and Jorge, hoping to uncover a familial link that would corroborate his suspicions. The plot thickens when he invites Jorge to Greg and Pam's engagement celebration, cleverly orchestrating an environment where Greg might inadvertently reveal the truth about their connection. However, when Greg vehemently denies any prior knowledge of Jorge, Jack remains skeptical and resorts to more drastic measures, administering a dose of truth serum that ultimately forces Greg into an involuntary confession: Pam is pregnant, and Jorge is his son. The revelation sends shockwaves through the partygoers, culminating in a stunned silence.

The next morning, Pam confronts Greg about Jorge, and despite her initial reservations, she chooses to believe him, willing to work through their issues together. Jack, however, has reached his breaking point, insisting that Pam and Dina accompany him as he departs the scene. Dina refuses to comply, revealing Jack's deceit by drugging Greg. As a result, the group turns against him, exposing the truth: everyone was aware of Pam's pregnancy, but kept it from Jack due to his inherent distrust. Devastated and hurt, Jack exits with Jorge in tow.

Bernie and Greg attempt to intercept Jack, only to be tasered and apprehended by Officer Vern LeFlore for speeding and failing to remain in Bernie's vehicle during the stop. Meanwhile, Jack receives the DNA test results, which reveal that Jorge's biological father is actually a baseball player who bears a striking resemblance to Greg. As he processes this new information, Jack turns back towards the house.

Witnessing Bernie and Greg being arrested, Jack tries to intervene on their behalf but ultimately ends up getting tasered and arrested alongside them by LeFlore.

Once released from custody by Judge Ira, a client of Roz's, Greg, Jack, and Bernie reunite. Before departing, Greg implores Jack and Bernie to put aside their animosity. Jack apologizes for his actions, sharing his CIA background with Bernie before expressing regret for his mistakes.

Greg and Pam tie the knot over the weekend, with Kevin, her former fiancé, now an ordained interfaith minister, officiating the ceremony. As Jack reviews hidden camera footage, he discovers each of the Fockers compromising the Ferber method in various ways, further solidifying the chaos that has engulfed their lives.
