She embarked on her acting career in 2003 following her graduation from Menlo School. O'Reilly initially appeared in a film entitled Bill the Intern and later featured in other films like Dinocroc, Nancy Drew, Just Add Water and Forgetting Sarah Marshall. She also exhibited versatility in her career by acting in television series like CSI: NY, Unhitched, The Vampire Diaries and Prime Suspect. In 2011, O'Reilly starred in The Help, a period piece set in 1960s Mississippi that earned rave reviews and box office success, grossing $211,608,112 globally and winning various ensemble awards. She subsequently featured in the 2013 film Jobs, a biography of technology pioneer Steve Jobs, co-starring Ashton Kutcher and Josh Gad. O'Reilly also demonstrated her talent in theatre by participating in the off-Broadway production titled The Robber Bridegroom by the Roundabout Theatre Company in 2016. A cast recording featuring her performance was released the same year. O'Reilly also played the lead role in the film Sleepwalker in 2017. Personally, she was romantically involved with actor James Franco before their break-up in 2011. As of 2021, she is married to writer and producer Dave Andron.
